An efficiency between 90% and 110% is generally considered acceptable for a reliable qPCR assay, corresponding to a slope roughly between -3.1 and -3.6.

Where does the slope value come from?

The slope comes from a standard curve: plotting the cycle threshold (Ct) value against the logarithm of starting template concentration across a dilution series, then fitting a straight line through the points.

What does an amplification factor of 2 mean?

An amplification factor of 2 means the amount of PCR product exactly doubles with every cycle, which is the theoretical maximum and corresponds to 100% efficiency.
