Why measure absorbance at 280nm for protein?
Aromatic amino acids, mainly tryptophan and tyrosine, absorb light strongly around 280 nanometers, making this wavelength a standard, convenient measure of total protein content in a sample.
How does the dilution factor get applied?
If a concentrated sample was diluted before its absorbance was measured, multiplying the calculated concentration by the dilution factor converts it back to the concentration of the original, undiluted stock sample.
