What is the 260nm absorbance conversion factor?
An absorbance of 1.0 at 260 nanometers corresponds to a known standard concentration for each nucleic acid type: 50 micrograms per milliliter for double-stranded DNA, 33 for single-stranded DNA, and 40 for RNA, since each molecule type absorbs 260nm light slightly differently.
Why does dilution factor matter?
If a sample was diluted before measuring its absorbance, multiplying by the dilution factor scales the measured concentration back up to the original, undiluted sample concentration.
Is this the same for synthetic oligonucleotides?
No, short synthetic oligonucleotide strands use a different, sequence-specific extinction coefficient and molecular weight rather than the standard conversion factors used here, since their absorbance properties depend heavily on their exact base sequence.
